BARI Special Training on Plant Tissue Culture & Directing BADC Kashimpur Tissue Culture Lab and Potato Cold Storage
By Md. Mamunur Reza (Assistant Program Director, APCU-BADC PARTNER & PhD Fellow)
Event Period: Specialized Training at BARI & Tenure as Assistant Director (TC), DD Office (TC) and Tissue Culture Laboratory, BADC Kashimpur (DEC 2022 – FEB 2023)
Clean, virus-free planting material is the foundation of modern horticultural productivity. Alongside completing intensive Special Training on Plant Tissue Culture at the Bangladesh Agricultural Research Institute (BARI), I served as Assistant Director (TC) at the DD Office (TC) and BADC Tissue Culture Laboratory in Kashimpur, Gazipur (টিসুকালচার এর উপর BARI এর বিশেষ প্রশিক্ষণ এবং BADC টিসুকালচার ল্যাব পরিচালনা).
Core Laboratory & Production Focus
- Meristem Culture & Virus Elimination: Producing pathogen-free nuclear stock for vegetatively propagated crops including potato, banana, strawberry, and ornamental orchids.
- Culture Media Optimization: Formulating Murashige and Skoog (MS) basal media with precise auxin-to-cytokinin ratios for rapid multiplication and vigorous rooting.
- Primary & Secondary Hardening: Managing the delicate transition of plantlets from 100% humidity culture vessels into sterile cocopeat plug trays inside climate-controlled mist chambers.
Potato Cold Storage & Breeder Seed Tuber Preservation (Kashimpur, Gazipur)
Alongside directing aseptic micropropagation in the BADC Tissue Culture Laboratory (Nov 2022 – Feb 2023), my mandate as Assistant Director (Tissue Culture Laboratory and Potato Cold Storage) included managing industrial cold-chain storage for tissue-culture-derived mini-tubers and foundation seed potatoes—maintaining strict temperature (2–4 °C), relative humidity (90–95%), and CO2 ventilation regimes to preserve tuber dormancy and vigor.
